Tymora?提供 pIMAGO——一種新穎的、非抗體的、高效且安全的產品,用于無偏地通用檢測任何磷酸化蛋白。

 

pIMAGO?基于水溶性的球狀納米聚合物(即樹枝狀大分子),其通過多個反應性基團(例如,Ti(IV)離子)進行多功能化,以高度特異性地識別磷酸基團,并通過“報告”基團(例如,生物素、熒光團)來促進其檢測。每個樹枝狀大分子的多個功能化基團確保了與磷酸化蛋白的非常強的結合(導致高特異性),并且來自許多“報告”基團的顯著信號放大(導致高靈敏度)。

基本原理

CRISPR-Cas9是一種細菌天然免疫系統,用于針對外源DNA進行有針對性的降解。它的工作機制是通過CRISPR RNA (crRNA) 和轉錄激活crRNA (trans-activating crRNA,tracrRNA) 之間的互補配對形成復合物,能夠特異性識別基因組中與其互補的序列。這種復合物可以引導Cas9內切酶切割目標DNA片段,導致DNA雙鏈斷裂的形成。這一過程實現了對目標基因組的精確編輯和修飾。如下圖所示:

通過基因工程手段對crRNA和tracrRNA進行改造,將其連接在一起得到sgRNA (single guide RNA)。通過將表達sgRNA的原件與表達Cas9的原件相連接,得到可以同時表達兩者的質粒,將其轉染細胞,便能夠對目的基因進行基因操作,在PAM (5’-NGG) 上游 ~3bp進行切割,如下圖所示:

因此本實驗的關鍵步驟是設計一對20bp(不包括NGG在內的)完全互補的oligo插入到如上圖所示的filler。另U6啟動子需要5’端的G起始轉錄,因此若設計的oligo第一個堿基非G,需額外增加一個G。

?詳細操作流程?

1. 設計sgRNA

確定靶基因的序列后,可通過在線網站設計(http://tools.genome-engineering.org ),或根據靶基因的CDS序列自行設計,最方便的是在human KO Library sgRNA里選擇。無論選擇哪種方式,都建議進行一下blast。

 

 

附上在線設計網頁截圖。輸入基因的name,填寫email address,設計結果稍后會發送到此郵箱,選擇序列的類型以及物種。如sequence type選擇unique region,一次只能輸入23~500bp的基因片段,最好一次只輸入一個外顯子,避免guide序列跨越內含子,都選擇鍵入后點擊提交,關注郵箱或者在線等。

分析中,顯示你所鍵入的基因序列里有36對可選的guide序列。

分析結束,點擊Download as genbank查看結果,此時也會收到郵件。

選擇分數較高的Guide序列,以Guide#1序列為例,2條單鏈oligo的序列如下:oligo1:5’-?caccGCGTTCAAGTACCAGTTCGTG -3’; oligo2:5’-?aaacCACGAACTGGTACTTGAACGc。標粗部分與經BsmBI酶切后的載體互補配對。BsmBI又名BbsI。

※oligoDNA序列的第一個堿基必須是G,如選取的Guide序列的第一個堿基不是G,需自行添加一個額外的G。

2. Oligo退火形成duplex

PCR儀 95℃ 5 min,緩慢降溫至室溫1h,1:200稀釋duplex。

3. 質粒酶切

4. 膠回收
大片段約11kb 回收;小片段約2kb為切下來的filler,不要。

5. 連接

室溫連接4-6 h。

6. 轉化(Amp+),挑克隆,搖菌,抽提質粒,測序!

7. 轉染

24孔板,細胞不要過滿,同實驗室日常轉染操作,可選擇性設置三個平行。同時轉染空載作為陰性對照。每孔500 ng。

8. puromycin篩選

轉染穩定48-72h后加puromycin進行篩選,1~3?g/mL,直至不再有細胞數量穩定,不再有細胞因不耐藥死去。

9. 單細胞分離

由于篇幅原因,有限稀釋法具體細節會在后面的文章單獨講解,請關注科研小助手后續文章。100個細胞鋪在一個大盤或96孔板中。剩余的細胞凍存。

10. 獲得單克隆細胞株

當肉眼可見類似于菌落的細胞團時,將其從大盤或96孔板消化下來轉移至12或24孔板中繼續擴大培養。注意不要污染到其它細胞團,不要選取過密的細胞團。

11. 擴大培養,提取細胞全基因組DNA和細胞總蛋白

12. 測序

Guide序列上游100bp左右和下游200bp左右為上下游設計一對引物,以全基因組為模板,以上述引物進行PCR。PCR產物送測序,或將產物用T7E1進行酶切消化檢測突變。

蛋白水解靶向嵌合體(PROteolysis TArgeting Chimeras, PROTACs)已成為靶向不可藥物化蛋白質組的新療法。PROTACs是同時與靶蛋白和泛素(Ub) E3連接酶結合的異雙功能小分子。這將會引誘目標蛋白的泛素化和降解。

?PROTACs作為一種新型成藥技術,可以讓藥物選擇性結合致病蛋白質,并通過“泛素-蛋白酶體”系統在細胞內自然降解,因而克服了傳統藥物抗藥性的問題。

PROTAC充當連接器

如圖所示,PROTAC充當連接器,幫助E3連接酶標記要降解的靶蛋白。整個過程就好像把你需要扔進垃圾桶的廢品打上一個便簽,這樣的話,凡是打上了便簽的靶蛋白,細胞統統都會處理掉。這也正是“泛素-蛋白酶體”系統的工作原理:給需要降解的靶蛋白貼一個“便簽”,然后降解掉。PROTAC無疑簡化了這個過程并極大的提高了效率。
